Wow, I stand corrected. Peter Pan's website does list Bourne Rotary as a stop. I ride my bicycle from the canal service road through the police barracks all the time to cross the bridge. I have never seen a bus stop at the Bourne Rotary. (please be patient and let the live webcam of the Bourne Rotary load)
I had trouble with Peter Pan's website. Where do you park to get the bus?
OP
IMHO, the visitors Center would be much easier and closer to Scusset to get a bus to Boston.

Across from Knights Inn is a convenience store. I don't know the details but next time you're there look in their parking lot. I think they're ok with day commuters but not overnight, but I really don't know.

I'm sure you're right. That must be a different bus, the Bourne bus is Woods Hole, Falmouth, Bourne rotary, South Station. So you can play the schedules off one against the other I suppose. Also, it probably makes sense for people staying at nearby Bay View Campgrounds.

I was able to get onto Peter Pan's website. I was wrong, but I see where the confusion is regarding the bus stop. The bus stop is called the Bourne Rotary stop, but is really down the street from the rotary. I take Freeman Street to the State Police Barracks from the Canal Service Road to cross the bridge. Freeman Street being closer to the rotary than the bus stop, I can not see the bus stop when making the trip.
OP
Should you decide to take a Peter Pan bus to Boston, cross the Bourne Bridge to the rotary, take your first exit from the rotary and stay straight (do not bear right). The bus stop is a little ways down on your right. A guess would be 6 to 7 miles and about 10 - 12 minutes from Scusset depending on traffic and lights.

We have taken that route before and find it easier than hitting Wash/Balt/NY and up through CT on 95. It is also less expensive--the only toll that you will have is crossing the Hudson until you get to MA/NH and ME.
The plan, for us, is New Green Acres in Walterboro SC, Walnut Hill KOA in Staunton VA, Promised Land State Park in Greentown PA and then on to New England.
This will be our 3rd year heading up. If any ?, PM me.
I took that same route last week from Fort Wilderness back home in MA. We hit 0 traffic and minimal construction. I stayed at New Green Acres as well. I would recommend Candy Hill Campground http://www.candyhill.com/ that was a really nice place to stay.
