Yes, the most direct route would be I95 south to SR 40 (Ormond Beach) , then west to Ocala.
2010 Flagstaff 26RLS
2012 Ford F-150 Screw Ecoboost H/D 3.73
2006 Toyota Tundra D/C (sold)
2001 Bantam B-19,(sold)
2003 Harley Road King
The whole family,
Cold ones in the cooler, good times ahead.

The interior routing that mdock2 suggested isn't bad but I would take I-95 over US1 any day, unless you like a lot of stop and go.

I would takethe I-295 west beltway to I-10 west to US301 south and take it all the way to SR40. Much easier than travelling the interstate. One warning, do not speed on US301 as their are numerous small towns that STRICTLY enforce the speed limit.
That works quite well from J'ville but not so much from St. Augustine. The two earlier routes both work well with the I-95/US40 being the easiest and probably about the same time even though farther.

You know, I didn't even see St Augustine in the title. My mistake

This poster has it nailed, except at Salt Spring take SR 314 to 40. All good roads, usually not much traffic.
If you are coming from the far south end of St. Augustine, take SR206 to 207, then etc, etc.

Absoultely correct on CR-314. This will cut many miles off of the trip. The road is not great but it is acceptable.
