I have a roll-up. When rolling up to use the 5er, I leave the back rail on, and with DW on one side and me on the other, we roll it up tightly and evenly. It's never been a problem.
There are a number of different models that will fold up completely opening the entire bed when folded up and attached over the rear window (Bak Flip is one that comes to mind) and my Extang would easily allow use of the forward third of the bed with the trailer attached. Lots of room to push things under the closed section but not as handy as the ones that fold up completely.

No problem on mine. I am sure something very light would fly out. But I put a ramp for the dog back there. And wood blocks for the front jacks have been left there with no problem.

Sure, but covering part of the bed helps reduce the lift. That's why I leave the cover closed as far as I can and still allow the pinbox to swing. The Vee tail gate helps too.
Just installed an Access Lorado rollup cover, which works our very well. Gives full access to entire bed, didn't cost as much as some of the fold-up covers that give full access, and has lifetime guarantee on the tarp. Very good option for $450.
I have the quad-fold on my long bed (installed by Chevy for the previous owner) and the trailer tows fine with all four pannels stacked in the forward position.
I installed a plywood partition across the bed under the forward hinge line and made the front compartment a lockable storage compartment for grill, chairs, extra power (50A) cord, soft drinks, etc.
By setting the rear pannels up in an inverted "V" under the nose of the trailer, I can open the front compartment while hitched.

Also have a tri-fold cover, wouldn't go anywhere without it. I once had a roll up cover that has a rail on each side that stuck up about 1 1/2 inches which lowered the distance from bottom of front of trailer to the rail. Went with the tri-fold, no rail system and much easier to fold back, maybe 5 seconds.
