All the advice here is great.
Couple things I would second:
* Never let your gas gauge go below 1/3. Great advice. Fill when you can to avoid the desperate "am I going to make it??" drive.
* Use Google maps to map out your route. With the street view you can get a good idea of what is available in particular towns. I have never done this, but if you are really worried you could completely map out your gas stops in this way. If your max fuel range on a full tank of gas is 250 miles then scan for an RV friendly fueling point every 200 miles.
* When stopping for food - do not attempt to tow your RV into a restaurant parking lot. You will get stuck and will end up backing out of a crowded lot. Look for restaurants where there is a super-market or empty parking lot within walking distance. Park in the empty lot and just walk to the restaurant. I once ended up backing out through a crowded Denny's lot. Total nail biter.
Finally - REMEMBER - you are on vacation! You do not need to try and drive 65-70 mph the entire way. No need to aggressively pass people or drive like a maniac. TAKE IT EASY. You will get better gas mileage and you will be much more relaxed when you arrive

Lake george is awesome... Stay to the highway and you should have no problem. You'll see a lot of TT so just follow the crowd. Check google earth and follow the highway and not the gas stations along the way sometime you can get a street view to see if you can pull off into the gas station. We stayed at schoonvalley.

This is a great thread I just came back from a 6 1/2 hour ride to williamsburg va as others have stated you will feel the push and pull of big rigs and buses just relax and slight tension on the wheel away from them, take your time a good set of mirrors makes a nice difference when making lane changes I usually put on my blinker and let it blink for about 10 to 15 seconds most cars driving beside me would speed up and alot of people would flash there high beams to let me know it was safe to enter there lane which I though was awesome I would then say thank you with a quick flick of my hazards. I found it helpful to stop every couple of hours and just walk and stretch I would also change my lumbar settings from time to time just to keep the seat feeling fresh. before heading out check the tire pressures and make sure the lugs are nice and snug and enjoy the trip

You've got a gas station on I87 north at mile marker 127 which should be roughly half way, can't get much easier then a highway gas stations. Depending on your route I hear there is construction at the GWB

I've used the Tappanzee heading out and back from NE a few times and had no issues, I just get in the center lane going west, and the right lane when heading east and into the tolls. You need to know where you want to be for the tolls way before you get there. EZpass is well worth getting if you don't already have it. You should know better then I but I try and get through the NYC/NJ area early in the am.

All good advice so far ...
I assume you will be taking the Thru Way up to Lake George ....its an easy ride. Keep in mind the rest stops are a little tight but you can fit just pay attention as you approach and plan your exit
Have fun in Lake Geoge we were there last month you will love it

Just take it slow. There's no harm in cruising along at 55-60 mph. The biggest issue is finding gas stations that are designed so you can get in and out without trouble. We start looking once we go below 1/2 tank so we can have time to find a good spot. Flying J and Petro are great. Many newer stations are fine too. We usually go in on one side, make a big loop near the store, and line up on the outside pump facing the road. This way we can just pull straight out. Sometimes you have to sit and wait for others to vacate the pumps, but that's just a price you pay to get the easy spot.
There's a free iPhone app called Road Ninja. It tells you what is at each exit.

The gas thing is the biggest thing I can really think of if you know the distance between point A and point B, try to figure out ahead of time where you can fill up. I have the same v10 in my f250 as your excursion and know that I am looking at around 8.5mpg towing our camper (it's heavy). When the computer on my truck tells me I have 100 miles left to go on the tank, I like to start looking for gas. That is if I have not planned on where to stop. I know that I can get a little over 250 miles on a tank of gas. So after about 150 miles of driving or every 2 hours, I want to stop and fill up. The only time I go beyond those marks is I haven't found a gas station I can get into yet, or I know where a specific one is within my range. Or I'm near my destination. The bigger the gas station, normally the easier to get into. We shoot for truck stops and travel stations.

Not sure if the gas stations near you vacuums the vapors while filling. I would turn off the refrigerator when gassing up. When the frig is on propane, you do have an open flame back there.
I agree about knowing your trailer height! The only true height is the one you measure yourself. Brochures vary in this area. I also was concerned about low clearances. If you have a Tom-Tom or Garmin GPS you shouold check out this link. I previously posted about it before, but, essentially, the POI Download saved our new TT.
