The burner on our stove in our RV can be light be turning a switch along the front of the stove, until it ignites, or you can light the burner with a match or lighter. However, the last time we were camping, it wouldn't ignite when turning the knob to the right two or three times. We have used it before and it worked the last time we were out camping. It did light with a lighter when we couldn't get it lite with the igniter switch.

Moisture maybe or low gas pressure or gas may not have reached the burner until you tried to light it.

Yea the igniter knob. 
Our old 86 King of the Road had a pilot light, that was sweet.
The igniter creates a spark at a location central to the three burners. Each burner has a tube that directs gas to the spark source. Either it is not creating a spark, or the tubes have been dislodged from the burner head.
I would guess no spark, check by lifting the top of the stove an look for it at the center of the three burner locations. You may need a new spark unit.

when you click the sparker, are you seeing the spark? I had this problem once and I lifted up the lid on the stove to expose the burner tubes and stuff and one of the sparker wires had come disconnected from that burner. plugged it back in and was back in business.

Lift the stove top and find where the sparker wires are connected. Pull these connections apart, wiggle them, clean the connections good, and then reconnect. Just a tiny bit of corrosion will prevent them from sparking at the burner. High humidity, like we have been having, is a real culprit. Had this all the time with the stove when I had a truck camper.

That thing is a little fickle for me. 50% chance it will light when I go there. I never looked into why, I just keep a lighter close

x3 on the igniter wires coming loose, as first suspicion. Keep matches/lighter handy, though. :-)

Might check the sides of your burners to. Sometimes a pot will boil over and clog the small gas openings in the side of the burner that feed the tubes to your central ignighter.

I adapted mine over to an electric sparker like most newer/bigger grills have. That puppy sparks like no tomorrow. Multiple discharges takes all the guesswork out of "is is sparking or not".
Theres a thread in the DIY forum somewhere that shows how to do it.

If the sparker is not producing a spark, look for an "AA" battery in the lighter wiring.
I just replaced a Piezo lighter on a BBQ grill. The button had the battery and the unit was sealed with 4 leads - 1 for each burner.
Be careful - they can produce a slight shock. Dont ask how I know
