I do not need to worry about DEF on my truck, but I am DEF illiterate. For those that do use it I am curious:
How much cost per gallon?
How much is used per each tank of diesel used?
Do you break it down in terms of MPG (for DEF) to calculate efficiency?
Does load or towing affect DEF efficiency?

2.5 gals @ WalMart costs ~$12.
Full tank of DEF (6+ gals.) lasts ~5000 miles (Truck in SIG)

And yes, the load or how hard you work the engine does effect the rate of consumption but not dramatically. Since DEF is injected into the exhaust stream to reduce NOx, the more fuel that is burned the more NOx is produced thus the more DEF is injected.
One of the interesting things about the addition of DEF systems is that the manufacturers can up the tuning parameters allowing the engine to produce more horsepower more efficiently while meeting EPA requirements.

I average 1000 miles per gallon towing a 12,000 lb 5th wheel. Use less when not towing. Cost is 2.79 per gallon at the bulk pumps, nearly twice that in the 2.5 gallon jugs.

Since it basically goes into the exhaust system,what happens if the tank goes empty? And if it still runs,why bother refilling it.Can't see the truck just shutting down.I can understand about the pollution stuff.

The EPA requires vehicle manufacturers to put measures in place to ensure that vehicles cannot run without DEF. Most manufacturers have approached this in a similar way. Before a truck's DEF tank runs empty the driver is given a series of alerts on their dashboard displays (much the same way as if they were running low on diesel). Generally speaking, when the DEF tank level drops below 10% an amber warning lamp will come on, at 5% this lamp starts flashing and below 2.5% a solid amber warning light is displayed.
If the truck is allowed to run out of DEF completely, the engine's power is reduced. This generally follows the next intentional key-off, when vehicle speed will be limited to 5 mph and a solid red warning will be displayed.

With Ford truck. As you use DEF you are alerted as to how many miles you can drive. Eventually the truck will not be able to operate at highway speed if you do not refill the tank and you will have to refill the DEF tank to get the truck to operate at highway speed. There are several alerts starting at 800 miles on your driver display telling you to refill your DEF tank. You can also select the DEF monitor in the driver display module to see how much DEF you have on board at any time.
Be aware that there are different qualities of DEF on the market. Check the specifications on the bottle.
