I'm thinking of buying a 2006 Jayco M29n travel trailer. My concern is the plumbing and gas lines in the slide-out. Has anyone had or know of any problems that have occurred because of often use of the slide-out. We plan to tour US next yr. and we will activate and de-activate slide quite often.

I have a kitchen slide in my motorhome. doesn't scare me a bit.
If it was a big problem, you can be sure that due to liability issues they wouldn't be making them anymore.

My Keystone has the sofa and kitchen on the slide. No problems yet, although it's less than a year old (but the slide has been in an out quite a few times already).

I for, for one, am bothered enough by the potential for problems that I would not buy one. But that's just me. Too many years spent fixing broken stuff. More things that flex, more possibility of breakage. Things that flex typically fail quicker than things that don't.
Is this backed up by real experience with a trailer with a slide like this? no.. Certainly you will hear from those that have them with no problems, but most that respond may only have a year or two on their TT. I wonder how many years it will take for the problems to start showing up?
Again, this is just me.. I don't like the toilet next to the bed either!

If I was concerned about mechanical failure I wouldn't own a slide unit in the first place. nor electric steps, etc. etc. etc.
the only safe thing would be to buy a small utility trailer and throw a mattress down on the floor.

My family and I have that exact unit except it is a 2005. We've begun our 4th year of camping where we do at least 2 weekends a month starting in May and ending in September. To date we've only had one problem with the kitchen connections and that had nothing to do with it being in the slider. Since all the plumbing is plastic the P-Trap fittings are only hand tight and I didn't check them one year and on the 3rd trip out it leaked. A quick twist and the problem was solved, now I check them a couple of times a season and no problems. I check the gas line to the stove a couple times a year but it doesn't show any type of wear at the flex points at all. We absolutely love having the kitchen in the slide (keeps down on the butt shuffle when we are both working on a meal). Also, having the kitchen & sofa in the slide means the dinette (and the big window in the dinette) faces into the campsite.

yep, that was a big concern of mine. I also like that big window on the curb side so I have improved vision to the side while driving/parking, etc.
