2-night minimums, especially for reservations, are common on weekends in our area. Most of the state parks, not to mention the private CGs, have such a rule during high season (June-Aug). Campinggrandma is right about the tourist-area hotels too.
And as one poster noted, the CG is likely full on weekends too. Any vacancies you see might very well be due to those who made reservations & didn't show up. In many CGs, those folks have already paid for the site, so the CG isn't really losing any money, though it may appear they have lost business.

Many hotels do, in the resort business. Which is comparable to destination RV parks (as opposed to motels and overnight RV parks along the highways).
The funny thing is these CG's want to complain if we stay overnight at Walmart, but they don't want us overnight! I can see their side to a point, but you would think when you're talking about one night within a week of the call, they would be able to make the call on availability.

I understand that CG's have certain policies, but to turn away business when you have openings is just plain nuts. Especially when in this economy CG's are not filling up like they used to. ...

Maybe it's a PA/northeast ecomony thing. Campgrounds here are full. If you don't have a reservation, you're not likely to find much.

This is an excellent point. If these campgrounds want to keep their business from going to the nearest Walmart lot, they had better start reserving a few sites for overnight stays.
Let's say they gave you the spot. They have now made an exception. Do you know what it's like when different folks pay different rates, or have to abide by different rules ?? Then the front desk REALLY has to pay. Or.....YOU abided by the 2 day rule and some guy next to you tells you that they made an exception for him. You would now be posting that the campground made an exception for someone and you're upset and will never step foot there again.

Sorry, but the two-day rule is just plain stupid when you have spaces available.
It might be their rule, but, it's a dumb rule.

It's a business policy I don't understand as well. It's not only confined to campgrounds. It seems some businesses rather make NO money than a little money. An empty site is generating no income. At least occupied at a lower cost is covering some of the fixed costs and cash flow. Like you said, it's just nuts.
